A DC Comics képregénye alapján készült széria egy lányról – Karáról (Melissa Benoist) – szól, aki a Krypton bolygón született és unokatestvéréhez – a híres Supermanhez – hasonló szuperképességekkel bír. A lányt a bolygó pusztulása előtt a Földre menekítették, ahol Superman talált Karának nevelőszülőket. A lány sikerrel titkolja képességeit és valódi kilétét, és teljesen átlagos életet él National Cityben, ahol a helyi újság nagyhatalmú tulajdonosának (Calista Flockhart) asszisztense. Kara a 24. születésnapján úgy dönt: ideje felfedni magát és azzá a hőssé lenni, akinek a sors szánta. Ebben segítségére lesz egy szupertitkos ügynökség vezetője (David Harewood) is, aki felkéri, hogy segítsen megvédeni a várost földönkívüli fenyegetésektől. A sorozat egyik érdekessége, hogy a készítők számos direkt és indirekt képregényes utalást csempésztek a szériába.
